The banding technique was applied in 1970 to study human metaphase chromosomes (a period of 320 bands), to describe previously unrecognized chromosomal abnormalities in many congenital malformations and neoplasms. The combination of banding technology and somatic hybridization helps to develop human gene mapping and is also a powerful stimulus for the study of chromosomal structure and evolution. In 1976, our laboratory synchronized cells with methotrexate and then exposed the cells to colchicine for a short period of time, thereby proposing techniques for performing routine studies on 400, 550, 850 and 1,200 band-era chromosomes.
