目的观察弓形虫感染对小鼠血清及睾丸的氧自由基和抗氧化酶水平的影响。方法将40只雄性BALB/c小鼠随机均分为4组,每组随机抽取5只腹腔注射弓形虫速殖子(2.5×103个/只,溶于0.2mlPBS),余5只为阴性对照,注射等量PBS。将各组小鼠分别于腹腔注射后第1、3、5和7天处死,取血和睾丸组织,检测血清和睾丸组织中一氧化氮(NO)、羟自由基(·OH)、抗超氧阴离子(O2-)自由基和超氧化物歧化酶(SOD)的含量。结果弓形虫感染组血清和睾丸组织中NO、·OH和O2-的浓度随时间延长逐渐升高,感染后第3、5和7天的浓度均显著高于对照组(P<0.01)。血清和睾丸组织内SOD浓度在感染后逐渐上升,第3天达峰值,后逐渐下降,感染后第3、5、7天与对照组差异有统计学意义(P<0.01)。结论弓形虫急性感染可引起小鼠血清及睾丸氧自由基(NO、·OH、O2-)和超氧化物歧化酶SOD水平的升高。
Objective To observe the effects of Toxoplasma gondii infection on the levels of oxygen free radicals and antioxidant enzymes in serum and testis of mice. Methods Forty male BALB / c mice were randomly divided into four groups. Toxoplasma tachyzoites (2.5 × 10 3 cells / mouse, 0.2 ml PBS) were randomly selected in each group, and the remaining 5 mice were used as negative control , Inject the same amount of PBS. The mice in each group were killed on the 1st, 3rd, 5th and 7th day after the intraperitoneal injection respectively. The blood and testicular tissues were taken for detection of the levels of nitric oxide (NO), hydroxyl radical (· OH) Oxygen anion (O2-) free radicals and superoxide dismutase (SOD) content. Results The concentrations of NO, · OH and O2- in serum and testicular tissue of Toxoplasma gondii infection group increased gradually with time. The concentrations of NO, · OH and O2- in serum and testis tissue were significantly higher than those in control group on the 3rd, 5th and 7th days after infection (P <0.01). The concentration of SOD in serum and testicular tissue increased gradually after infection, reaching the peak on the third day and then decreasing gradually. There was a significant difference (P <0.01) on the 3rd, 5th and 7th day after infection with the control group. Conclusion The acute infection of Toxoplasma gondii can cause the elevation of serum free oxygen and free radicals (NO, · OH, O2-) and superoxide dismutase (SOD) in mice serum and testis.
