目的:检测常氧与模拟海拔4 000 m低氧条件下大鼠睾丸组织中死亡受体5(DR5)和FLIP抑制蛋白(c-FLIP)表达水平,以及c-FLIP在睾丸组织中的分布。方法:40只10周龄雄性SD大鼠随机分为常氧组,低氧3、15、30 d组。低氧各组分别置模拟海拔4 000 m低压舱中减压3、15、30 d;常氧组在平原(海拔300 m)喂养;减压结束后取睾丸组织,Western印迹检测睾丸组织中DR5与c-FLIP蛋白表达,免疫荧光法检测睾丸组织中c-FLIP分布。结果:低氧3、15、30 d组大鼠睾丸DR5表达量分别为2.04±0.11、1.97±0.12、2.34±0.11,均显著高于常氧组DR5的表达(1.78±0.09,P均<0.05)。低氧15、30 d组大鼠睾丸c-FLIP表达量分别为0.87±0.03、0.74±0.07,均显著低于常氧组c-FLIP的表达(1.03±0.02,P均<0.05)。结论:模拟海拔4 000 m低氧促进睾丸组织凋亡蛋白DR5表达,抑制睾丸组织抗凋亡蛋白c-FLIP表达。
OBJECTIVE: To detect the expression of DR5 and FLIP inhibitor protein (c-FLIP) and the distribution of c-FLIP in testis tissue under normoxia and simulated hypoxia in 4 000 m altitude rats. Methods: Forty male 10-week-old SD rats were randomly divided into normoxia group, hypoxia group, The hypoxia groups were set to simulate the hypoxia at 4 000 m altitude for 3, 15 and 30 days respectively. The normoxia group was fed on the plain (300 m above sea level). After the end of decompression, the testis tissue was taken and Western blot was used to detect the DR5 And c-FLIP protein expression, immunofluorescence test c-FLIP distribution in testis. Results: The expression of DR5 in testis of hypoxia group was 2.04 ± 0.11, 1.97 ± 0.12 and 2.34 ± 0.11 respectively, which were significantly higher than that of normoxic group (1.78 ± 0.09, P <0.05 respectively) ). The expression of c-FLIP in testis of hypoxia group was 0.87 ± 0.03,0.74 ± 0.07, which were significantly lower than that of normoxic group (1.03 ± 0.02, P <0.05). Conclusion: Simulating hypoxia at 4 000 m altitude can promote the expression of DR5 protein in testis tissue and inhibit the expression of anti-apoptotic protein c-FLIP in testis tissue.
