目的:分析宁波地区2008年麻疹病毒H和N基因的序列特征。方法:采用RT-PCR扩增2008年宁波地区麻疹分离株血凝素蛋白(H)和核蛋白(N)C末端456 bp核苷酸并进行序列测定,与GenBank中麻疹病毒各基因型参考株比较并分析毒株变异情况。结果:共分离到11株麻疹病毒,均为H1a基因亚型。与H1a参考株China93-2的H和N基因的氨基酸同源性分别为98.5%~98.7%和96.1%~98.7%;与中国疫苗株S191的H和N基因的氨基酸同源性分别为95.0%~95.5%和86.8%~89.5%。结论:宁波地区2008年流行的麻疹病毒均为H1a基因亚型,与中国疫苗株S191在基因特征上存在明显差异。
Objective: To analyze the sequence characteristics of H and N genes of measles virus in Ningbo in 2008. Methods: The 456 bp nucleotide sequence of hemagglutinin (H) and nucleocapsid (N) at the C terminus of measles isolates from Ningbo in 2008 was amplified by RT-PCR and sequenced. The results were compared with the reference strains of genotypes of measles virus in GenBank Compare and analyze strain variation. Results: 11 strains of measles virus were isolated, all of which were H1 subtype. The amino acid homologies of the H and N genes of Chinese strain H1_3 with the Chinese strain H1_93 were 98.5% -98.7% and 96.1% -98.7%, respectively. The amino acid homologies with the H and N genes of Chinese vaccine strain S191 were 95.0% ~ 95.5% and 86.8% ~ 89.5%. Conclusion: The majority of measles viruses in Ningbo in 2008 were all H1a genotypes, which were significantly different from those of Chinese vaccine strain S191.
