本工作在大鼠异丙肾上腺素(ISO)心肌坏死模型上,首次观察内皮素A 受体拮抗剂BQ123的治疗的作用。结果表明,ISO 心肌坏死动物血浆内皮素(ET)放免活性明显升高;BQ123显著抑制ISO 引起的血浆丙二醛(MDA)、心肌MDA 和钙含量的升高,减轻其病理过程;但对ISO 引起的心功能抑制和组织损伤无明显改善。实验结果提示,ET 可能是ISO 心肌损伤发病因素之一;应用ET 特异拮抗剂对于缺血性心脏疾病可能具有辅助防治作用。
This work in the rat isoproterenol (ISO) myocardial necrosis model, the first observation of the endothelin A receptor antagonist BQ123 treatment. The results showed that the plasma endothelin (ET) radioimmunoassay activity was significantly increased in ISO myocardial necrosis rats. BQ123 significantly inhibited the increase of plasma malondialdehyde (MDA), myocardial MDA and calcium levels caused by ISO, and reduced the pathological process; but ISO Caused by cardiac dysfunction and tissue damage without significant improvement. The experimental results suggest that ET may be one of the risk factors of myocardial injury in ISO. The use of ET-specific antagonists may be helpful in the prevention and treatment of ischemic heart disease.
