目的:制备雄黄微生物炮制液,观察其药效和毒性的变化。方法:利用氧化亚铁硫杆菌制备雄黄炮制液;以常规炮制雄黄作为对照,以人多发性骨髓瘤细胞株RPMI8226为细胞模型,应用噻唑蓝(MTT)法和流式细胞仪(FCM),检测两种药物对细胞增殖与细胞周期的影响;以离子发射原子光谱仪法(ICP-AES),测定其中砷在小鼠血液和组织中分布的浓度变化。结果:与常规炮制雄黄相比,雄黄微生物炮制后,溶解度提高,用药剂量显著降低,对细胞抑制凋亡作用增强,蓄积量显著降低,在大鼠各组织器官内几乎未检测出砷的分布。结论:微生物新法炮制雄黄后,药效增强、砷蓄积毒性降低,为科学炮制雄黄提供了新的思路。
Objective: To prepare the realgar liquid and observe the change of its efficacy and toxicity. Methods: Realgar preparation was prepared by using Thiobacillus ferrooxidans. Real-time processing of realgar was used as a control. The human multiple myeloma RPMI8226 cell line was used as a cell model. MTT assay and flow cytometry (FCM) The effects of two drugs on cell proliferation and cell cycle were studied. The concentrations of arsenic in blood and tissues of mice were measured by ICP-AES. Results: Compared with conventional realgar, the realgar had a higher solubility and a lower dose of realgar. The inhibition of apoptosis was enhanced and the stock volume was significantly reduced. Almost no distribution of arsenic was detected in tissues and organs of rats. CONCLUSION: After the micro-organism was processed into realgar, its efficacy was enhanced and its arsenic accumulation toxicity was reduced. It provided a new idea for scientific preparation of realgar.
