目的 研究叶酸治疗对胃癌前病变组织中bcl 2、bax及p5 3基因表达的影响。方法 胃镜活检经病理证实为胃癌前病变患者 3 8例 ,利用逆转录聚合酶链式反应 (RT PCR)方法检测胃癌前病变组织bcl 2、bax基因表达率 ,利用流式细胞仪检测组织p5 3蛋白表达率。将患者随机分为治疗组 (叶酸 10mg ,每天三次 )与对照组 (硫糖铝 1.0 ,每天四次 )各 19例 ,治疗结束复查组织中bcl 2 ,bax基因表达率及p5 3蛋白表达率。结果 治疗组治疗后bcl 2基因表达率降低 (P <0 .0 5 ) ,bax基因表达率无明显变化 (P >0 .0 5 ) ,p5 3蛋白表达率增高 (P <0 .0 5 ) ,对照组治疗后各项指标无明显变化 (P >0 .0 5 )。结论 叶酸干预可促进胃癌前病变组织中p5 3基因的表达 ,抑制bcl 2基因的表达 ,而对bax基因表达无明显影响
Objective To study the effect of folic acid treatment on the expression of bcl-2, bax and p53 in gastric precancerous lesions. Methods 38 cases of gastric precancerous lesions confirmed by pathological biopsy were enrolled in this study. The expression of bcl-2 and bax gene in gastric precancerous lesions was detected by reverse transcriptase-polymerase chain reaction (RT-PCR). The expression of p53 was detected by flow cytometry Protein expression rate. The patients were randomly divided into treatment group (folic acid 10mg, three times a day) and control group (sucralfate 1.0, four times a day each) 19 cases. At the end of treatment, the expression rates of bcl 2 and bax and the expression of p5 3 protein were observed. Results The expression of bcl-2 gene in the treatment group decreased (P <0.05), the expression of bax gene had no significant change (P> 0.05) and the expression of p5 3 protein increased (P <0.05) There was no significant change in the indexes of the control group after treatment (P> 0.05). Conclusion Folic acid intervention can promote the expression of p5 3 gene and inhibit the expression of bcl 2 gene in gastric precancerous lesions without affecting the expression of bax gene
