目的观察何首乌主要成分二苯乙烯苷(THSG)对去卵巢肾虚大鼠骨髓间充质干细胞(MSCs)增殖及旁分泌细胞因子mRNA的影响。方法切除SD雌性大鼠双侧卵巢复制肾虚大鼠模型,密度梯度离心法收集各组大鼠MSCs进行体外培养,观察THSG对大鼠第3代MSCs增殖及旁分泌细胞因子mRNA的影响。结果模型组大鼠第3代MSCs增殖的OD值明显低于正常组(P<0.05,P<0.01),THSG组明显高于模型组(P<0.05)。模型组MSCs表达粒-巨噬细胞集落刺激因子(GM-CSF)、白血病抑制因子(LIF)、粒细胞集落刺激因子(G-CSF)、骨形成蛋白-2(BMP-2)mRNA明显低于正常组(P<0.05);THSG组MSCs表达G-CSF mRNA明显高于模型组(P<0.05)。结论与正常大鼠比较,去卵巢大鼠MSCs增殖能力减弱,可能与其MSCs旁分泌GM-CSF、LIF、G-CSF、BMP-2 mRNA减少有关。THSG改善去卵巢肾虚大鼠MSCs增殖能力,可能与其促进MSCs表达G-CSF mRNA有关。
Objective To observe the effect of stilbene glucoside (THSG), a major component of Polygonum multiflorum Thunb. On proliferation and paracrine cytokines mRNA of bone marrow mesenchymal stem cells (MSCs) of ovariectomized rats. Methods SD rat ovariectomized rat model of kidney deficiency was excised. MSCs of each group were collected and cultured in vitro by density gradient centrifugation. The effects of THSG on the proliferation and paracrine cytokines mRNA of 3rd generation MSCs were observed. Results The proliferation of 3rd generation MSCs in model group was significantly lower than that in normal group (P <0.05, P <0.01). The THSG group was significantly higher than that in model group (P <0.05). The expression of granulocyte-macrophage colony-stimulating factor (GM-CSF), leukemia inhibitory factor (LIF), granulocyte colony stimulating factor (G-CSF) and bone morphogenetic protein-2 (BMP- (P <0.05). The expression of G-CSF mRNA in THSG group was significantly higher than that in model group (P <0.05). Conclusion Compared with normal rats, the proliferation of MSCs in ovariectomized rats is weakened, which may be related to the decrease of paracrine expression of GM-CSF, LIF, G-CSF and BMP-2. THSG can improve the proliferation of MSCs in ovariectomized rats, which may be related to its effect on the expression of G-CSF mRNA in MSCs.
