本文报导用萤光法测定盐酸普鲁卡因酰胺的含量。本法系基于试样在 pH7.5的缓冲液中,与 Flu-orescamine 试剂反应生成萤光团(Huorophore),在激发及发射波长分别为400及485毫微米处测定其萤光强度。在0.04~1微克/毫升的浓度范围内呈线性关系(r=0.999),并至少在二小时内稳定。本法准确度高、重复性好,总回收率达99.8%,标准偏差为±1.14(n=5)。本法与美国药典第18版的测定法比较,具有简便、快速及灵敏度高等优点。
This article reports the determination of procaine hydrochloride by fluorimetry. This method is based on the reaction of a sample with Flu-orescamine reagent in a buffer of pH 7.5 to generate a fluorophore (Huorophore) whose fluorescence intensity is measured at excitation and emission wavelengths of 400 and 485 nm, respectively. The results showed a linear relationship (r = 0.999) in the range of 0.04 to 1 μg / ml and was stable for at least two hours. This method has high accuracy and repeatability, with a total recovery of 99.8% and a standard deviation of ± 1.14 (n = 5). This method and the United States Pharmacopoeia 18th edition of the determination method, with the advantages of simple, rapid and high sensitivity.
