目的 :建立血清中茶碱的提取和薄层色谱扫描定性和定量检测方法。方法 :血清用 1%盐酸酸化 ,乙醚 二氯甲烷 (6∶4)或氯仿萃取 ,点于GF2 54 薄层板上 ,正庚烷∶三氯甲烷∶无水乙醇∶乙酸乙酯∶甲醇∶冰乙酸 (5∶2∶2∶1.5∶2 .5∶0 .5 )展开 ;薄层扫描检测。结果 :茶碱薄层层析Rf值为 0 .35 ;最大吸收波长为 2 75nm ;血中茶碱线性检测范围为 1~ 2 4μg/ml,最小检出浓度为 0 .4μg/ml。 结论 :本法快速、简便、定性定量准确 ,可用于茶碱中毒的快速诊断和体内药物浓度监测。
Objective: To establish a qualitative and quantitative method for the extraction of theophylline in serum and TLC scanning. Methods: The serum was acidified with 1% hydrochloric acid, extracted with ether (dichloromethane) (6: 4) or chloroform, spotted on a GF2 54 plate, n-heptane: chloroform: absolute ethanol: ethyl acetate: methanol: Acetic acid (5: 2: 2: 1.5: 2 .5: 0 .5); thin layer scan detection. Results: The Rf value of theophylline TLC was 0.35. The maximum absorption wavelength was 2 75 nm. The linearity of theophylline in blood was 1-24 μg / ml and the minimum detectable concentration was 0.4 μg / ml. Conclusion: The method is rapid, simple, qualitative and quantitative, and can be used for the rapid diagnosis of theophylline poisoning and the monitoring of the drug concentration in the body.
