MYB转录因子参与植物对外界逆境胁迫反应的调控,为提高水稻的耐盐性,本研究对水稻OsMYB56基因进行了生物信息学分析,利用RT-PCR技术克隆了水稻耐盐碱基因OsMYB56,构建了植物表达载体pCAMBIA3300-Os MYB56,并通过农杆菌介导法将OsMYB56基因导入水稻中,以进一步研究其功能。对转化植株进行PCR、RT-PCR、Southern杂交及Bar蛋白试纸条检测,证明OsMYB56基因已整合到水稻基因组中。本研究所获得的转基因水稻材料对水稻耐盐碱育种具有重要的意义。
In order to improve the salt tolerance of rice, the MYB transcription factor is involved in the regulation of plant stress response to external environment. In this study, bioinformatics analysis of rice OsMYB56 gene was carried out. The salt tolerance gene OsMYB56 was cloned by RT-PCR and constructed The plant expression vector pCAMBIA3300-Os MYB56 was transformed into rice by Agrobacterium tumefaciens-mediated method to further study its function. Transgenic plants were subjected to PCR, RT-PCR, Southern blotting and Bar protein test strips to confirm that the OsMYB56 gene has been integrated into the rice genome. The transgenic rice materials obtained in this study are of great significance for salt-tolerant breeding in rice.
