免疫毒素(IT)杀伤试验分8组进行。主要采用改进的MTT法测定不同浓度的抗CD3-PAPs、抗CD7-PAPs、抗CD3-PAPS十抗CD7—PAPs、多抗AHTG—PAPs杀伤T细胞的效率,同时以B细胞作非靶细胞对照。结果证实,两种抗人T细胞单抗IT和多抗IT均具有特异性杀伤T细胞的作用。比较杀伤率发现,单抗IT特异性高,杀伤力也较强;多抗IT杀伤力强,但特异性较差;而两种单抗IT合用则杀伤力进一步提高,并能保持其特异性。结果提示,我们制备的两种单抗IT可用于体夕P清除T细胞作异体骨髓和/或脐带血移植;两者联合应用可能其协同作用效果更好。
Immunotoxin (IT) killing test was divided into 8 groups. The efficiency of anti-CD3-PAPs, anti-CD7-PAPs, anti-CD3-PAPS ten anti-CD7-PAPs and multi-anti-AHTG-PAPs killing T cells were tested by modified MTT assay. . The results confirmed that both anti-human T cell monoclonal antibody IT and multi-anti-IT have a specific anti-T cell role. Compared with the killing rate, it was found that the monoclonal antibody has high IT specificity and high lethality, while the multiple anti-IT has strong lethality, but the specificity is poor. However, the combination of the two monoclonal antibodies IT further increases its lethality and maintains its specificity. The results suggest that the two monoclonal antibodies we prepared IT can be used to eliminate T cells for allogeneic bone marrow and / or cord blood transplantation; the combination of the two may be better synergistic effect.
