目的使用卵清蛋白经口灌胃致敏不同周龄的BN大鼠,初步探讨哺乳期暴露致敏蛋白对BN大鼠致敏性影响,并了解血清中细胞因子变化。方法选2周龄、4周龄和8周的BN大鼠,经口灌胃1 mg/kg·bw·d剂量卵清蛋白,连续35 d。第28天和第35天时内眦静脉采血,采用酶联免疫吸附法测定血清中OVA特异性Ig E(s Ig E)抗体、细胞因子IL-4、IL-10和IFN-γ浓度。结果第28天时,8周龄组BN大鼠血清中OVA s Ig E浓度显著高于对照组(P<0.05),2周龄和4周龄组与对照组之间差异无统计学意义(P>0.05)。第35天时,4周龄组和8周龄组大鼠血清中OVA s Ig E浓度都显著高于对照组(P<0.05),2周龄组与对照组比较差异无统计学意义。同时第35天细胞因子检测结果,2周龄组BN大鼠血清中IL-10和IFN-γ浓度降低(P<0.01),IL-4浓度变化不大。4周龄组和8周龄组BN大鼠血清中IL-4和IL-10浓度增加(P<0.05),血清中IFN-γ浓度变化不大。结论哺乳期会抑制卵清蛋白致敏BN大鼠,这可能受到细胞因子影响。
Objective To orally inoculate ovalbumin (OVA) into BN rats of different weeks of age to study the allergenicity of sensitized protein exposed during lactation to BN rats and to understand the changes of serum cytokines. Methods BN rats at 2 weeks, 4 weeks and 8 weeks were orally administered orally with 1 mg / kg · bw · d orally for 35 days. Serum samples were collected on day 28 and day 35. Serum levels of OVA-specific IgE (anti-IgE) antibodies, cytokines IL-4, IL-10 and IFN-γ were measured by enzyme-linked immunosorbent assay. Results On the 28th day, the serum concentrations of OVA s Ig E in BN rats of 8-week-old group were significantly higher than those of the control group (P <0.05), but there was no significant difference between the 2-week-old and 4-week-old rats and the control group > 0.05). On the 35th day, the serum concentrations of OVA s Ig E in 4-week-old and 8-week-old rats were significantly higher than those in the control group (P <0.05). There was no significant difference between the 2-week-old and the control groups. Meanwhile, on the 35th day, the concentrations of IL-10 and IFN-γ in the serum of BN rats decreased (P <0.01) and the concentration of IL-4 changed little in the 2-week-old BN rats. The levels of IL-4 and IL-10 in serum of 4-week-old and 8-week-old BN rats increased (P <0.05), while the concentration of IFN-γ in serum did not change much. Conclusion Lactation inhibits ovalbumin-sensitized BN rats, which may be influenced by cytokines.
