目的 观察慢性激怒应激对大鼠血及结肠粘膜中前列腺素 E2 (PGE2 )、 c AMP、 c GMP含量的影响 ,探讨其作用和意义。方法 大鼠用夹子将其后肢夹住 ,直立位挂起 ,并让大鼠之间出现扭打嘶咬。钳夹时间第 1天为 30 min,以后每天增加 10 min,连续 2周。以不作任何处理的大鼠作正常对照。造模结束后取血浆及结肠粘膜 ,以放免法测定血及结肠粘膜中 PGE2 、 c AMP、c GMP含量。结果 大鼠血中 PGE2 、c AMP、c GMP含量和 c AMP/ c GMP与正常对照组之间无明显差异 ;大鼠结肠粘膜中 PGE2 、c AMP的含量及 c AMP/ c GMP较正常组明显升高 (P<0 .0 5 ,P<0 .0 1) ;而 c GMP的含量则与正常组之间无显著差异。结论 慢性激怒应激可引起大鼠结肠粘膜 PGE2 和环核苷酸的变化 ,这些变化可能在胃肠功能紊乱性疾病中起一定的作用。
Objective To investigate the effect of chronic irritation stress on prostaglandin E2 (PGE2), cAMP, cGMP in blood and colonic mucosa of rats and its significance and significance. Methods Rats were clamped with their hindlimbs, the upright position was suspended, and writhing and bite between the rats occurred. The first day of clamping time was 30 min, and then increased by 10 min every day for 2 weeks. Rats without any treatment were used as normal control. Plasma and colon mucosa were taken after modeling, and the contents of PGE2, c AMP and c GMP in blood and colonic mucosa were determined by radioimmunoassay. Results The levels of PGE2, cAMP, cGMP and cAMP / c GMP in rat blood were not significantly different from those in normal control group. The levels of PGE2 and cAMP and cAMP / c GMP in colonic mucosa were significantly higher than those in normal group (P <0.05, P <0.01), while the content of cGMP had no significant difference with normal group. Conclusion Chronic irritant stress can cause changes of PGE2 and cyclic nucleotide in colonic mucosa of rats, and these changes may play a role in gastrointestinal disorders.
