本文报道一例人脑多形性胶质母细胞瘤细胞系“BT325”的建立及其生物学特性,至今已传180代,维持4年以上,生长稳定,每周传代一次,细胞形态多样,星球状,双极形,星形及少量多核巨细胞。透射电镜可见多量微管及束状微丝。异种移植至裸鼠皮下生长肿瘤,病理切片经 HE、PTAH 及 Foot’s 染色均证实为恶性胶质瘤。免疫化学染色胶质纤维酸性蛋白(GFAP)呈阳性反应,生长曲线计算倍增时间为36.5小时,染色体组型显示亚四倍体,90%以上显示有标记染色体。细胞周期时相分析用FCM 法计算出各时相的百分比为 G_1=52.94%,S=28.08%,G_2+M=18.98%,用 PLM 法计算出各时相的时间为 T_c=37.5小时、TG_1=16.2小时,T_s=10.5小时,TG_2+M=10.8小时,TM 估计值为0.8小时,细胞冻融存活率为77%左右,认为本系细胞生长和特性稳定,可作筛选化疗药物,有一定帮助。
This article reports a human glioblastoma multiforme cell line “BT325” established and its biological characteristics, has been transmitted 180 generations, maintaining more than 4 years, stable growth, once a week passage, cell morphology, the planet Like, bipolar, star and a small amount of multinucleated giant cells. Cell cycle phase analysis calculated by FCM percentage of each phase as G_1 = 52.94%, S = 28.08%, G_2 + M = 18.98%, calculated by PLM method for each phase time T_c = 37.5 hours, TG_1 = 16.2 hours, T_s = 10.5 hours, TG_2 + M = 10.8 hours, TM estimated value of 0.8 hours, cell freezing and thawing survival rate of about 77%, that the Department of cell growth and characteristics of stability, can be used for screening chemotherapeutic drugs, there is a certain help.
