本文在已克隆9.1C3抗体VH和Vk基因的基础上,用一编码疏水性多肽接头的DNA,经重叠延伸拼接PCR,构建了9.1C3单链抗体基因。经测序表明,VH、Vk及Linker的序列均正确,为9.1C3单链抗体的表达奠定了基础。
Based on the cloned VH and Vk genes of 9.1C3 antibody, the 9.1C3 single-chain antibody gene was constructed by overlap extension splicing PCR using a DNA encoding a hydrophobic polypeptide linker. Sequencing showed that the sequences of VH, Vk and Linker were correct, which laid the foundation for the expression of 9.1C3 single chain antibody.
