本文在已克隆９．１Ｃ３抗体ＶＨ和Ｖｋ基因的基础上，用一编码疏水性多肽接头的ＤＮＡ，经重叠延伸拼接ＰＣＲ，构建了９．１Ｃ３单链抗体基因。经测序表明，ＶＨ、Ｖｋ及Ｌｉｎｋｅｒ的序列均正确，为９．１Ｃ３单链抗体的表达奠定了基础。
Based on the cloned VH and Vk genes of 9.1C3 antibody, the 9.1C3 single-chain antibody gene was constructed by overlap extension splicing PCR using a DNA encoding a hydrophobic polypeptide linker. Sequencing showed that the sequences of VH, Vk and Linker were correct, which laid the foundation for the expression of 9.1C3 single chain antibody.
