利用葡聚糖凝胶(DG)首次特异地减慢水杨酸钠(SS)从胰岛素直肠栓剂(INS)中的释放速度,而不影响胰岛素(IN)的释放,从而达到两者同步吸收的作用。试验证明,含200mg大交联度DG即DGG-10的INS与不含SG-10的INS相比能明显增加INS中IN在免直肠中的吸收量,致使兔血浆中IN最大浓度不变的情况下,持续时间明显延长,进而血溏浓度下降持续时间延长。INS中含400mgDGG-10反而使血浆中IN浓度和血糖浓度的变化程度小于不含DGG-10INS,分析可能是过量的DGG-10过度地减慢了SS从INS中的释放速度,以至于SS始终不能达到促进IN吸收的浓度的结果。
