目的观察波动高糖和持续高糖对人脐静脉内皮细胞(HUVEC)增殖及凋亡的影响。方法以体外培养HUVEC为研究对象,实验分为3组:正常组(5.5mmol·L-1)、持续高糖组(30.5 mmol·L-1)、波动高糖组(每24 h轮换加入5.5mmol·L-1或30.5 mmol·L-1葡萄糖溶液)。用CCK-8法和流式细胞仪,分别检测细胞存活率及凋亡率,倒置相差显微镜观察细胞形态。结果持续及波动高糖均可明显抑制HUVEC增殖,诱导其凋亡,与正常组比较各组细胞间差异有统计学意义(P<0.05)。其中波动高糖条件下细胞生存率又低于持续高糖,凋亡率则相反。结论高浓度葡萄糖能抑制培养的HUVEC增殖,促进细胞凋亡,且波动高糖较持续高糖对血管内皮细胞可能具有更强的损伤效应。
Objective To observe the effects of fluctuating high glucose and continuous high glucose on the proliferation and apoptosis of human umbilical vein endothelial cells (HUVECs). Methods HUVECs were cultured in vitro. The experiment was divided into three groups: normal group (5.5mmol·L-1), continuous high glucose group (30.5 mmol·L-1), fluctuating high glucose group mmol·L-1 or 30.5 mmol·L-1 glucose solution). Cell viability and apoptosis rate were detected by CCK-8 assay and flow cytometry respectively. Cell morphology was observed by inverted phase contrast microscope. Results Both continuous and fluctuating high glucose could significantly inhibit the proliferation and induce apoptosis of HUVECs. Compared with the normal group, there was significant difference between the groups (P <0.05). One of the conditions under fluctuating high glucose cell survival rate was lower than sustained high glucose, the rate of apoptosis is the opposite. Conclusion High concentration glucose can inhibit the proliferation of cultured HUVECs and promote the apoptosis of HUVECs. The high level of glucose may have more damaging effects on vascular endothelial cells than the continuous high glucose.
