目的研究利用组织培养技术快速获得独一味组培苗的方法。方法以野生独一味为外植体,MS为基本培养基,研究不同浓度激素对叶片愈伤组织诱导及分化的影响。结果适宜野生独一味叶片愈伤组织诱导的培养基为MS+NAA0.2mg/L+2.4-D 1.0mg/L+6-BA 2.0mg/L;适宜不定芽分化的培养基为MS+6-BA 0.5mg/L+NAA 0.05mg/L;适宜生根培养基为MS+IBA 0.5mg/L+NAA 0.1mg/L。结论通过研究组织培养中不同激素配比,获得了独一味组培苗,初步建立了独一味快繁技术。
Objective To study the method of using Tissue culture technique to obtain Tissue culture seedlings of Duyiwei. Methods Wild single flavor as explants, MS as a basic medium to study the different concentrations of hormones on leaf callus induction and differentiation. The results showed that the medium suitable for the treatment of wild single leaves was MS + NAA 0.2mg / L + 2.4-D 1.0mg / L + 6-BA 2.0mg / L. The medium suitable for adventitious bud differentiation was MS + 6- BA 0.5mg / L + NAA 0.05mg / L; suitable rooting medium for MS + IBA 0.5mg / L + NAA 0.1mg / L. Conclusion Through the study of different hormones in tissue culture, Tissue culture seedlings were obtained, and the unique and rapid propagation technology was established.
