目的观察血小板源性生长因子(PDGF)和转化生长因子β1(TGF-β1)对大鼠肺成纤维细胞整合素(integrin)α5β1及其配体纤维连接蛋白(FN)合成的影响。方法采用细胞培养、Northern印迹杂交、免疫细胞化学方法。结果经PDGF-BB、TGF-β1作用2h,α5、β1、FNmRNA的表达即强于对照组,以后继续增强,其间仅于18或12h短暂回落,但仍明显强于对照组。免疫细胞化学显示肺成纤维细胞α5β1和FN蛋白的表达也明显增强。结论PDGF、TGF-β1可以促进integrinα5β1及其配体FN的合成增加,其调控机制之一可能是在转录水平上增强integrinα5β1及其配体FNmRNA的表达。
Objective To observe the effects of platelet derived growth factor (PDGF) and transforming growth factor β1 (TGF-β1) on the synthesis of integrin α5β1 and its ligand fibronectin (FN) in rat lung fibroblasts. Methods Cell culture, Northern blot hybridization and immunocytochemistry were used. Results The expression of α5, β1 and FNmRNA in PDGF-BB and TGF-β1 groups was stronger than that in control group after 2 and 10 h of TGF-β1 treatment, and then continued to increase after a short period of 18 or 12 h, but still significantly stronger than that of control group. Immunocytochemistry showed that the expression of α5β1 and FN protein in lung fibroblasts was also significantly increased. Conclusion PDGF and TGF-β1 can promote the synthesis of integrinα5β1 and its ligand FN. One of the possible mechanisms is that the expression of integrinα5β1 and its ligand FN mRNA may be increased at the transcriptional level.
