目的:优化超声辅助提取石莼多糖的工艺条件及评价石莼多糖清除DPPH·自由基的能力。方法:超声辅助提取石莼多糖,在单因素试验基础上,利用正交试验进行工艺优化。结果:超声辅助提取石莼多糖的最优条件为:液料比50∶1、超声时间40 min、超声温度70℃、pH 8.0。此优化条件下,石莼多糖得率为28.07%,清除DPPH·自由基的半数抑制率(IC50)为0.87 mg/mL。结论:超声辅助提取石莼多糖提取率高、重复性好,可作为提取石莼多糖的一种工艺方法,为将石莼多糖开发为一种抗氧化剂奠定基础。
OBJECTIVE: To optimize the ultrasonic extraction conditions of polysaccharides and to evaluate the ability of polysaccharides to scavenge DPPH · free radicals. Methods: Ultrasound - assisted extraction of polysaccharides from Lithocarpus japonicus, on the basis of single factor experiment, using orthogonal experiment to optimize the process. Results: The optimum conditions of polysaccharides extraction by ultrasonic were as follows: the ratio of material to material was 50:1, the time of ultrasonic was 40 min, the temperature of ultrasonic was 70 ℃, pH 8.0. Under the optimum conditions, the yield of polysaccharide from Lithocarpus japonicus was 28.07%, and the half-value inhibition (IC50) of scavenging DPPH · radical was 0.87 mg / mL. CONCLUSION: Ultrasound-assisted extraction of polysaccharide from Lithocarpus japonicus has high extraction rate and good repeatability, which can be used as a technological method to extract polysaccharides from Lithocarpus polystachyus, laying a foundation for the development of polysaccharides from Lithocarpus as an antioxidant.
