目的研究维吾尔族代谢综合征(MS)患者chemerin在腹部大网膜、皮下脂肪组织的基因表达及其血清蛋白水平的变化,并探讨其变化在MS中的可能作用。方法选取120例慢性胆囊炎和非胆囊炎维吾尔族患者,在排除了慢性胆囊炎对临床生化指标和MS可能的影响后,选择81例慢性胆囊炎择期手术患者作为chemerin研究的对象,并将其分为MS组(39例)和非MS组(42例)。用realtimeRT-PCR方法检测两组脂肪组织chemerinmRNA的表达水平,ELISA法测血浆chemerin浓度,并测量腰围、臀围、舒张压、收缩压、空腹血糖和血脂等。结果①MS组血浆chemerin水平显著高于非MS组(P<0.01);大网膜和皮下脂肪组织chemerinmRNA的表达水平差异在两组之间均无统计学意义。②血浆chemerin水平与皮下脂肪组织chemerinmRNA表达量呈正相关(r=0.469,P=0.001),与年龄、三酰甘油、腰臀围比及胰岛素抵抗指数呈正相关(r=0.434,P<0.01;r=0.368,P=0.003;r=0.251,P=0.031;r=0.225,P=0.041)。结论维吾尔族MS患者血浆chemerin水平显著升高,且与MS的几个关键指标相关,可能在维吾尔人MS的发生发展中起作用。
Objective To investigate the gene expression of chemerin in the omental omentum and subcutaneous adipose tissue in patients with metabolic syndrome (MS) in Uygur and its serum protein levels and to explore the possible role of chemerin in MS. Methods A total of 120 Uygur patients with chronic cholecystitis and non-cholecystitis were enrolled in the study. After eliminating the possible impact of chronic cholecystitis on clinical biochemical parameters and MS, 81 patients with chronic cholecystitis elective surgery were selected as the targets of chemerin study. Divided into MS group (39 cases) and non-MS group (42 cases). The expression of chemerin mRNA in adipose tissue was detected by realtime RT-PCR. The concentration of chemerin in plasma was measured by ELISA. The waist circumference, hip circumference, diastolic blood pressure, systolic blood pressure, fasting blood glucose and blood lipids were measured. Results ① The level of chemerin in MS group was significantly higher than that in non-MS group (P <0.01). There was no significant difference in chemerin mRNA expression between omentum and subcutaneous adipose tissue between the two groups. ② The level of chemerin in plasma was positively correlated with the expression of chemerin mRNA in subcutaneous adipose tissue (r = 0.469, P = 0.001), and positively correlated with age, triglyceride, waist-hip ratio and insulin resistance index = 0.368, P = 0.003; r = 0.251, P = 0.031; r = 0.225, P = 0.041). Conclusions The plasma levels of chemerin in Uygur MS patients are significantly increased, and are related to several key indicators of MS, which may play a role in the occurrence and development of MS in Uygur.
