背景:淫羊藿是中医治疗骨质疏+松症常用中草药中使用频率最高者之一,但对其抗骨质疏松有效成分的研究尚不多见。目的:研究淫羊藿总黄酮提取物(TFE)的抗骨质疏松活性。设计:完全随机,前后和组间对照实验研究。地点和对象:在解放军兰州军区兰州总医院动物实验科完成,清洁级SD大鼠48只,由甘肃省医学科学研究院实验动物中心提供。干预:48只大鼠分为6组,行双侧卵巢切除术OVX或假手术。3组行()OVX者分别以50,100和200mg/(kg·d)的剂量灌服TFE,1组行OVX者灌服1.5mg/(kg·d)的尼尔雌醇作为阳性对照。3个月后处死所有大鼠。主要观察指标:术后大鼠血尿生化指标、骨组织形态学、骨密度和股骨力学强度检查。结果:OVX导致骨微结构退化、骨量丢失和股骨力学强度下降。OVX组血清ALP为219.41±34.52)U/L,显著高于正常对照组;TFE200和(尼尔雌醇组分别为(164.32±37.96)和(150.30±44.83)U/L,显著低于OVX组;血清OC浓度变化与ALP相似。尿Ca/Cr,P/Cr和DPD/Cr水平OVX组均显著高于正常对照组,尼尔雌醇组各项指标均显著低于OVX组t=4.17~4.50,P<0.05)。OVX组股骨骨密度显著低于正常(对照组,TFE200和尼尔雌醇组显著高于OVX组。L2~L4腰椎变化与股骨相似。灌服TFE抑制或减弱了OVX的作用,尤其200mg/(kg·d)的疗效与尼尔雌醇相当,但
BACKGROUND: Epimedium is one of the most commonly used herbal medicines for the treatment of osteoporosis and osteoporosis in traditional Chinese medicine, but the study of its anti-osteoporosis active ingredients is rare. Objective: To study the anti-osteoporosis activity of total flavonoids extract (TFE) of Epimedium. Design: Completely random, controlled experimental studies before and after and between groups. Sites and objects: Completed in the Animal Experiment Department of Lanzhou General Hospital of the Lanzhou Military Command of the Chinese People’s Liberation Army. 48 clean SD rats were provided by the Experimental Animal Center of the Gansu Academy of Medical Sciences. Intervention: 48 rats were divided into 6 groups and were subjected to bilateral OVX or sham surgery. Three groups of OVX were fed with TFE at doses of 50, 100 and 200 mg/(kg·d), respectively. One group of OVX was fed with 1.5 mg/(kg·d) of nilestriol as a positive control. All rats were sacrificed 3 months later. MAIN OUTCOME MEASURES: Biochemical indicators, bone histomorphology, bone density, and mechanical strength of femurs were examined in postoperative rats. RESULTS: OVX resulted in degraded bone microarchitecture, loss of bone mass, and decreased femur mechanical strength. Serum ALP in OVX group was 219.41±34.52)U/L, which was significantly higher than that in normal control group; TFE200 and (Nilestriol group were (164.32±37.96) and (150.30±44.83) U/L, respectively, significantly lower than OVX group. The change of serum OC concentration was similar to that of ALP, urinary Ca/Cr, P/Cr and DPD/Cr levels in OVX group were significantly higher than those in normal control group, and nilestriol group had significantly lower indexes in OVX group than t=4.17~ 4.50, P<0.05).
