本实验用尼龙毛将经聚蔗糖—泛影葡胺分离的PBL分为粘附与非粘附部分,尔后分别用Et_(AET)花结—酯酶、C_3花结—酯酶两种双标记法分别测定,进一步分析酯酶在淋巴细胞中的分布.取正常肝素抗凝血4~5ml,用淋巴细胞分层液提出淋巴细胞,再以尼龙毛过滤,过滤柱应用结核菌素注射器,装填洁净的尼龙毛70mg,柱高6cm,以10%小牛血清—Hank’s液作营养洗液,收获的非粘附细胞主要为T细胞,粘附细胞主要为B细胞.前者用Et_(AET)—酯酶双标法测定,后者用C_3花结—酯酶双标法测定.
In this experiment, PBLs separated by Ficoll-diatrizoic acid were separated into two parts: adhesion and non-adhesion, and then double-labeled with Et_ (AET) -acetate and C_3- Method to determine the distribution of esterase in lymphocytes.Use normal heparin anticoagulant blood 4 ~ 5ml, with lymphocyte stratified liquid proposed lymphocytes, and then nylon wool filtration, filtration column using tuberculin syringe, filling Clean nylon wool 70mg, column height 6cm, with 10% calf serum-Hank’s solution as a nutritional lotion, harvested non-adherent cells mainly T cells, adherent cells mainly B cells.The former with Et_ (AET) - Esterase double standard method, the latter with C_3 flower knot - esterase double standard method.
