目的研究白藜芦醇对高糖条件下系膜细胞增殖的影响,为白藜芦醇治疗糖尿病肾病提供实验依据。方法体外培养大鼠系膜细胞,分为5组:对照组、高糖组、白藜芦醇2.5、5、10μmmol/L组;应用细胞计数K it8(CCK8)法测定细胞增殖,流式细胞仪检测细胞周期和细胞凋亡。结果高糖组细胞增殖率为(105.74±1.78)%,较对照组增高(P<0.01);白藜芦醇组细胞增殖率分别为(101.38±3.62)%、(96.55±1.28)%、(91.95±2.40)%,较高糖组减低(P<0.05或P<0.01);高糖组G1期细胞比例(62.86±1.57)%较对照组(57.93±2.70)%增高(P<0.05),白藜芦醇组G1期细胞比例较高糖组减少,其中10μmmol/L组与高糖组比较差异有统计学意义(P<0.05);高糖组S期细胞比例(29.00±1.08)%较对照组(34.91±1.43)%减少(P<0.01),白藜芦醇组S期细胞比例较高糖组增高,其中5、10μmmol/L组与高糖组比较差异有统计学意义(P<0.01);白藜芦醇5、10μmmol/L组细胞凋亡率分别为(2.20±0.53)%、(4.53±0.31)%,明显高于高糖组(P<0.01)。结论白藜芦醇能抑制高糖诱导的系膜细胞增殖;白藜芦醇抑制增殖与影响细胞周期及诱导凋亡有关。
Objective To study the effect of resveratrol on the proliferation of mesangial cells under high glucose conditions and provide experimental evidence for the treatment of diabetic nephropathy with resveratrol. Methods Rat mesangial cells were cultured and divided into five groups: control group, high glucose group, resveratrol 2.5, 5, 10μmmol / L group; cell proliferation was measured by K8 (CCK8) Instrument to detect cell cycle and apoptosis. Results The proliferation rate of cells in high glucose group was (105.74 ± 1.78)%, which was higher than that in control group (P <0.01). The cell proliferation rate in resveratrol group was (101.38 ± 3.62)%, (96.55 ± 1.28)% (P <0.05 or P <0.01). The percentage of cells in G1 phase of high glucose group (62.86 ± 1.57)% was higher than that in control group (57.93 ± 2.70)% (P <0.05) The percentage of G1 phase cells in resveratrol group was lower than that in high glucose group, and the difference was significant between 10μmmol / L group and high glucose group (P <0.05). In high glucose group, the proportion of cells in S phase was (29.00 ± 1.08)% Compared with the control group (34.91 ± 1.43)% (P <0.01), the percentage of S phase cells in the resveratrol group was higher than that in the high glucose group, and there was significant difference between the 5 and 10μmmol / L group and the high glucose group (P < 0.01). The apoptotic rates of resveratrol in 5 and 10μmmol / L groups were (2.20 ± 0.53)% and (4.53 ± 0.31)%, respectively, which were significantly higher than those in high glucose group (P <0.01). Conclusion Resveratrol can inhibit the proliferation of mesangial cells induced by high glucose. Resveratrol inhibits the proliferation and affects cell cycle and induces apoptosis.
