描述用乙酸α—生育酚作内标剂同时测定血清视黄醇和α—生育酚的高效液相色谱法。视黄醇、α—生育酚和内标剂的相对保留时间分别为2.60、5.56和7.14分钟,峰形锐利、对称。方法的回收率视黄醇平均97.2%,α—生育酚为95.6%。方法的精密度:视黄醇批内CV2.0—4.2%(=1.41—3.99μmol/L),批间 CV5.3%(=1.50μmol/L);α—生育酚批内和批间 CV 分别为0.4~2.4%(=15.2—50.4μmol/L)及3.9%(=15.4μmol/L)。视黄醇和α—生育酚的线性范围分别为0.44—6.98和3.86—6.166μmol/L。用本法测得正常人血清视黄醇浓度男性为2.33±0.58μmol/L,女性为1.62±0.38μmol/L;血清α—生育酚男性为16.32±4.81μmol/L,女性为14.70±4.27μmol/L。
Description High performance liquid chromatography for the simultaneous determination of serum retinol and alpha-tocopherol using alpha-tocopheryl acetate as an internal standard. Retinol, α-tocopherol and internal standard relative retention time of 2.60,5.56 and 7.14 minutes, peak shape sharp, symmetrical. The recovery of the method was 97.2% retinol average and 95.6% alpha-tocopherol. The precision of the method was CV2.0-4.2% (= 1.41-3.99μmol / L) for retinol and CV5.3% (= 1.50μmol / L) They were 0.4-2.4% (= 15.2-50.4μmol / L) and 3.9% (= 15.4μmol / L), respectively. The linear range of retinol and alpha-tocopherol ranged from 0.44 to 6.98 and from 3.86 to 6.166 μmol / L, respectively. The normal serum retinol concentration in this method was 2.33 ± 0.58μmol / L for males and 1.62 ± 0.38μmol / L for females and 16.32 ± 4.81μmol / L for serum α-tocopherol and 14.70 ± 4.27μmol for females respectively / L.
