用体外短期液体培养技术,观察RA和不同来源的造血调控因子对HL-60细胞增殖与分化的影响。以生长曲线和倍增时间作为观察各种试剂对HL-60细胞增殖作用的指标;以NBT还原反应和细胞形态变化作为分化成熟的指标。结果是RA、PHA-LCM单用时对HL-60细胞均有抑制增殖和诱导分化作用,PWM-SCM、LP3-CM仅部分抑制增殖,而FGF、Hu-CSF和EPO等无效。各种造血生长因子分别与RA合用时,能加强RA对HL-60细胞的增殖抑制和诱导分化作用的有FCF、PWM-SCM、PHA-LCM和Hu-CSF等;LP3-CM仅增强抑制增殖:EPO未显示作用。
Using in vitro short-term liquid culture techniques, the effects of RA and hematopoietic regulators from different sources on the proliferation and differentiation of HL-60 cells were observed. The growth curve and doubling time were used as indicators of the proliferation of HL-60 cells observed with various reagents, and NBT reduction and cell morphology were used as indicators of differentiation maturation. As a result, both RA and PHA-LCM inhibited proliferation and induced differentiation of HL-60 cells when used alone. PWM-SCM and LP3-CM only partially inhibited proliferation, whereas FGF, Hu-CSF and EPO were ineffective. When hematopoietic growth factors are combined with RA, they can enhance the inhibition of proliferation and induction of differentiation of HL-60 cells by FCF, PWM-SCM, PHA-LCM and Hu-CSF; LP3-CM only enhances proliferation inhibition. : EPO does not show effect.
