目的研究长药隔重楼Paris polyphylla var.pseudothibetica根茎中的化学成分,为阐明其有效成分及扩大重楼属植物的药用资源提供科学依据。方法利用正相硅胶柱色谱、葡聚糖凝胶Sephadex LH-20、正反相制备色谱等手段进行分离纯化,并通过1H-NMR、13C-NMR、ESI-MS等波谱技术进行结构鉴定。结果从长药隔重楼中分离得到了12个化合物,分别鉴定为:薯蓣皂苷元(1)、薯蓣皂苷元-3-O-α-L-呋喃阿拉伯糖基-(1→4)-[α-L-吡喃鼠李糖基-(1→2)]-β-D-吡喃葡萄糖苷(2)、偏诺皂苷元-3-O-α-L-呋喃阿拉伯糖基-(1→4)-β-D-吡喃葡萄糖苷(3)、偏诺皂苷元-3-O-α-L-呋喃阿拉伯糖基-(1→4)-[α-L-吡喃鼠李糖基-(1→2)]-β-D-吡喃葡萄糖苷(4)、偏诺皂苷元-3-O-α-L-吡喃鼠李糖基-(1→4)-α-L-吡喃鼠李糖基-(1→4)-[α-L-吡喃鼠李糖基-(1→2)]-β-D-吡喃葡萄糖苷(5)、β-谷甾醇(6)、豆甾醇(7)、胡萝卜苷(8)、豆甾醇-3-O-β-D-吡喃葡萄糖苷(9)、山柰酚(10)、β-蜕皮激素(11)、蔗糖(12)。结论化合物1~9为首次从该植物中分离得到。
Aim To study the chemical constituents in the rhizomes of Paris polyphylla var.pseudothibetica and provide a scientific basis for clarifying the active ingredients and expanding the medicinal resources of the plants belonging to the genus Rheum. Methods The constituents were isolated and purified by normal phase silica gel column chromatography, Sephadex LH-20 dextran gel and reversed-phase preparative chromatography. The structures were identified by 1H-NMR, 13C-NMR and ESI-MS spectroscopy. Results Twelve compounds were isolated from the spacer of the long-chain drug-eluting Fraxinus mandshurica respectively and identified as diosgenin (1), diosgenin -3-O-α-L-arabinofuranosyl- (1 → 4) α-L-rhamnopyranosyl- (1 → 2)] - β-D-glucopyranoside (2) → 4) -β-D-glucopyranoside (3), sapogenin-3-O-α-L-arabinofuranosyl- (1 → 4) (1 → 2)] - β-D-glucopyranoside (4), sapogenin-3-O-α-L-rhamnopyranosyl- - Pyran rhamnosyl- (1 → 4) - [α-L-rhamnopyranosyl- (1 → 2)] - β-D-glucopyranoside (5) 6), stigmasterol (7), daucosterol (8), stigmasterol-3-O-β-D-glucopyranoside (9), kaempferol (10), β-ecdysteroid (11) (12). Conclusion Compounds 1 to 9 were isolated from this plant for the first time.
