目的:观察辛夷挥发油体外对大鼠胸腔炎性白细胞脂氧酶(5-LO)活性的影响,探讨其干预花生四烯酸代谢的抗炎机制。方法:以角叉菜胶所致急性胸膜炎模型大鼠的胸腔炎性白细胞为基本反应体系,分别加入CaCl2、MgCl2、花生四烯酸(AA)、A23187以及辛夷挥发油(0.011、0.044、0.088、0.132mg/ml)或齐留通(0.625μg/ml),提取完整白细胞的5-LO代谢产物白三烯B4(LTB4)和5-羟基二十碳四烯酸(5-HETE),运用反相高效液相色谱(RP-HPLC)紫外检测分离测定LTB4和5-HETE的合成水平,来反映5-LO的活性。结果:辛夷挥发油在0.011~0.132mg/ml浓度范围内,对大鼠胸腔白细胞LTB4与5-HETE的生物合成呈现抑制趋势。结论:辛夷挥发油体外可抑制大鼠胸腔白细胞花生四烯酸代谢酶5-LO的活性,降低LTB4与5-HETE合成水平,提示其抗炎作用可能与抑制5-LO活性,减少致炎代谢产物的生成有关。
Objective: To observe the effects of volatile oil of Magnolia biondii on the activity of lipoxygenase (5-LO) in rats with thoracic inflammatory cytokines in vitro and to explore its anti-inflammatory mechanism of intervention on the metabolism of arachidonic acid. Methods: Thoracic inflammatory leukocytes from rat model of acute pleuritis induced by carrageenan were used as basic reaction system. CaCl2, MgCl2, arachidonic acid (AA), A23187 and volatile oil of Magnolia biondii (0.011,0.044,0.088,0.132 (LTB4) and 5-hydroxy eicosatetraenoic acid (5-HETE) were extracted from intact leukocytes by using a reversed phase High-performance liquid chromatography (RP-HPLC) UV detection of LTB4 and 5-HETE separation level of synthesis to reflect the 5-LO activity. Results: The volatile oil of Magnolia biondii in the concentration range of 0.011 ~ 0.132mg / ml inhibited the biosynthesis of LTB4 and 5-HETE in rat pleural leukocytes. Conclusion: Volatile oil of Magnolia biondii can inhibit the activity of arachidonic acid metabolizing enzyme 5-LO and reduce the synthesis of LTB4 and 5-HETE in rat pleural leukocytes in vitro, suggesting that its anti-inflammatory activity may be related to inhibition of 5-LO activity and reduction of inflammatory metabolites The generation of.
