目的:建立测定大鼠血浆中N-去甲氧羰基代谢物(DCJW)的高效液相色谱法。方法:以乙腈为沉淀剂处理大鼠血浆后,用反相高效液相色谱-紫外法测定。色谱柱为Shim-pack VP-ODS(150 mm×4.6 mm,5μm);柱温:室温;流动相为乙腈-水(80∶20);流速1.0 mL·min-1;检测波长314 nm。结果:N-去甲氧羰基代谢物(DCJW)的线性范围为0.025~5.00μg·mL-1,r=0.9998;最低检测限为0.020μg·mL-1。血浆中DCJW的绝对回收率为97.0%~100.5%,方法回收率为95.2%~101.0%,日内和日间RSD均小于8.0%。结论:本法专属性强,灵敏度高,重复性好,适用于DCJW血药浓度测定及药代动力学研究。
Objective: To establish a HPLC method for the determination of N-demethoxycarbonyl metabolites (DCJW) in rat plasma. Methods: The plasma of rats was treated with acetonitrile as precipitant and determined by reversed-phase high performance liquid chromatography-ultraviolet spectrophotometry. The column was Shim-pack VP-ODS (150 mm × 4.6 mm, 5 μm). The column temperature was at room temperature and the mobile phase was acetonitrile-water (80:20). The flow rate was 1.0 mL · min-1. Results: The linear range of N-demethoxycarbonyl metabolites (DCJW) was 0.025-5.00μg · mL-1, r = 0.9998. The lowest detection limit was 0.020μg · mL-1. The absolute recovery rate of plasma DCJW was 97.0% ~ 100.5%. The recovery rate of the method was 95.2% ~ 101.0%. The intra-day and inter-day RSD were less than 8.0%. Conclusion: This method is specific, sensitive and reproducible. It is suitable for the determination of DCJW blood concentration and pharmacokinetics.
