目的观察小剂量阿糖胞苷对HL 6 0细胞生长与分化的影响及其可能的作用机制。方法阿糖胞苷 (10 8M)作用HL 6 0细胞 2 6天 ,观察细胞生长 ,用流式细胞仪分析细胞周期 ,用免疫组化法检测P5 3、P2 1蛋白表达的变化 ,原位杂交法检测P5 3mRNA表达水平的改变。结果小剂量阿糖胞苷可诱导HL 6 0细胞向单核 /巨噬细胞分化 ,抑制HL 6 0细胞的增殖 ,使细胞停滞于G1期 ,P5 3,P2 1蛋白和P5 3mRNA表达明显增强。结论小剂量阿糖胞苷对HL 6 0细胞具增殖抑制和诱导分化作用 ,该作用与G1期阻滞及P5 3,P2 1表达增高有关。
Objective To observe the effect of low dose of cytarabine on the growth and differentiation of HL-60 cells and its possible mechanism. Methods Cytosine (10 8M) was used to effect HL 6 0 cells for 26 days. The cell growth was observed. The cell cycle was analyzed by flow cytometry. The expression of P53 and P2 1 was detected by immunohistochemistry. Method to detect P5 3 mRNA expression level changes. Results Low doses of cytarabine induced the differentiation of HL-60 cells into monocytes / macrophages, inhibited the proliferation of HL-60 cells, and arrested cells in G1 phase. The expressions of P53, P2 1 and P53 mRNA were significantly increased. Conclusion Low-dose cytarabine can inhibit the proliferation and induce the differentiation of HL-60 cells, which is related to the arrest of G1 phase and the increase of P5 3 and P2 1.
