目的:探究磷脂酰肌醇-3激酶δ的表达与妊娠期糖尿病(GDM)的相关性,为临床治疗GDM提供理论依据。方法:选取2012年1月~2013年7月在河北医科大学第二医院行产前检查的孕妇166例,其中正常孕妇44例为NGT组、GDM孕妇122例为GDM组,测量并记录两组孕妇年龄、身高、体重、孕周,并计算BMI指数;采集孕妇的血液样品并收集血液中的白细胞,通过生化实验检测血糖、血脂、空腹胰岛素(FIN)、胆固醇、C反应蛋白(CRP)、高密度脂蛋白(HDL)等指标水平,并计算稳态模型β细胞功能指数(HOMA-B)以及胰岛素抵抗指数(HOMA-IR);采用RT-PCR方法检测PIK3CD mRNA在白细胞内的表达水平。结果:两组孕妇年龄、身高、体重、孕周、BMI值等指标比较,差异无统计学意义(P>0.05),GDM组HDL-C含量明显低于NGT组,空腹1 h和2 h后的葡萄糖浓度(FPG)及糖化血红蛋白(HbA1C)含量均明显高于NGT组,两组比较,差异有统计学意义(P<0.05);两组血甘油三酯(TG)、低密度脂蛋白胆固醇(LDL-C)、血清总胆固醇(TC)、CRP、HOMA-IR以及HOMA-B等指标比较,差异无统计学意义(P>0.05)。GDM组白细胞内的PIK3CD mRNA是NGT组的1.92倍。结论:GDM患者白细胞内的PIK3CD过量表达可引起HDL-C含量减少,并导致血糖浓度增加。
Objective: To investigate the relationship between the expression of phosphatidylinositol 3-kinase (kD) and gestational diabetes mellitus (GDM), and to provide a theoretical basis for clinical treatment of GDM. METHODS: A total of 166 pregnant women undergoing prenatal examinations at the Second Hospital of Hebei Medical University from January 2012 to July 2013 were selected. Among them, 44 normal pregnant women were NGT group and 122 GDM pregnant women were GDM group. Two groups of pregnant women were measured and recorded The pregnant women’s age, height, weight and gestational age were calculated and the BMI index was calculated. Blood samples of pregnant women were collected and leukocytes were collected. Blood glucose, lipids, fasting insulin (FIN), cholesterol, C-reactive protein (HOMA-B) and insulin resistance index (HOMA-IR) were calculated. The expression of PIK3CD mRNA in leukocytes was detected by RT-PCR. Results: There was no significant difference in the age, height, weight, gestational age and BMI between two groups (P> 0.05). The content of HDL-C in GDM group was significantly lower than that in NGT group at 1 and 2 h after fasting (P <0.05). The levels of triglyceride (TG), low density lipoprotein cholesterol (LDL-C) and total cholesterol in blood serum of two groups were significantly higher than those of NGT group (LDL-C), serum total cholesterol (TC), CRP, HOMA-IR and HOMA-B. There was no significant difference between the two groups (P> 0.05). The PIK3CD mRNA in GDM group was 1.92 times higher than that in NGT group. CONCLUSION: Overexpression of PIK3CD in leukocytes of GDM patients results in a decrease of HDL-C content and an increase of blood glucose concentration.
