目的优化延胡索有效成分的提取、纯化工艺。方法以提取物中延胡索乙素量为考察指标,在单因素实验基础上,采用正交试验的方法对影响延胡索有效成分提取、纯化的因素进行优化。结果最佳提取工艺为:药材粗粉(10目)于60℃加12倍量水浸泡3次,每次30 min;最佳纯化工艺为:提取液用药材1.5倍量的WLD大孔吸附树脂吸附,3倍树脂量的70%乙醇洗脱。结论优化的提取、纯化工艺简单、稳定且能有效保证延胡索乙素不被破坏。
Objective To optimize the extraction and purification of the active ingredient of Corydalis yanhusuo. Methods The content of tetrahydropalmatine in the extract was taken as the index of investigation. On the basis of the single factor experiment, orthogonal test was used to optimize the factors affecting the extraction and purification of the active ingredient of Corydalis. Results The optimum extraction process was as follows: the crude meal (10 mesh) was soaked in 12 times of water at 60 ℃ for 30 min each time; the best purification process was as follows: the WLD macroporous adsorption resin Adsorption, 3 times the amount of resin 70% ethanol elution. Conclusion Optimized extraction, purification process is simple, stable and can effectively ensure that tetrahydropalmatine is not destroyed.
