目的:建立荧光定量RT-PCR法检测肺癌组织中人类有机阴离子转运多肽(OATP)B和DmRNA的表达,并分析其表达与病理分型及临床资料的关系。方法:SYBRGreen实时荧光定量PCR定量检测OATP-B和OATP-D的mRNA在40例肺癌中的表达,分析其在不同病理分型肺癌和正常肺组织间及有无淋巴结转移肺癌间的表达差异,并用10例正常肺组织作对照。结果:与正常肺组织相比,OATP-B和OATP-D的mRNA在肺癌中表达均上调,差异有统计学意义,P<0.05;OATP-B和OATP-D的mRNA在有无淋巴结转移肺癌中的表达差异有统计学意义,P<0.05。结论:肺癌中OATP-B和OATP-D的mRNA表达均明显高于正常肺组织,且OATP-B和OATP-D的mRNA在淋巴结转移肺癌中的表达均高于无淋巴结转移肺癌。
OBJECTIVE: To establish a fluorescent quantitative RT-PCR method to detect the expression of human organic anion transport polypeptide (OATP) B and DmRNA in lung cancer and to analyze its relationship with pathological classification and clinical data. Methods: The mRNA expressions of OATP-B and OATP-D in 40 lung cancer tissues were detected by SYBR Green real-time quantitative PCR. The expression of OATP-B and OATP-D mRNA in lung cancer tissues was detected by SYBR Green quantitative real- Ten normal lung tissues were used as controls. Results: OATP-B mRNA and OATP-D mRNA were up-regulated in lung cancer compared with normal lung tissue, the difference was statistically significant, P <0.05; OATP-B and OATP-D mRNA in lung cancer with or without lymph node metastasis The difference was statistically significant, P <0.05. CONCLUSIONS: OATP-B and OATP-D mRNA expressions in lung cancer were significantly higher than those in normal lung tissues, and the expression of OATP-B and OATP-D mRNA in lung cancer with lymph node metastasis were higher than those without lymph node metastasis.
