目的研究人端粒酶逆转录酶对胶质瘤细胞系体外生物学行为的影响,以期寻找控制胶质瘤细胞恶性行为的有效途径。方法应用小分子RNA干扰技术抑制T98G胶质瘤细胞系中端粒酶逆转录酶的表达,用MTT法、TUNEL法、Transwell分别检测T98G胶质瘤细胞中人端粒酶逆转录酶被抑制后对细胞体外增殖能力、细胞凋亡、细胞侵袭、迁移能力等生物学行为的影响。结果人端粒酶逆转录酶小分子RNA干扰片段能够有效抑制酶活性,抑制T98G胶质瘤细胞增殖(P<0.05)、诱导细胞凋亡(P<0.05)并抑制细胞的迁移与侵袭(P<0.05)。结论通过抑制人端粒酶逆转录酶的活性能够有效抑制胶质瘤细胞的体外恶性生物学行为,促进细胞凋亡。
Objective To study the effect of human telomerase reverse transcriptase on the biological behavior of glioma cell lines in vitro in order to find an effective way to control the malignant behavior of glioma cells. Methods The expression of telomerase reverse transcriptase in T98G glioma cell lines was inhibited by small interfering RNA interference technique. Human telomerase reverse transcriptase was detected by MTT assay, TUNEL assay and Transwell assay in T98G glioma cells On cell proliferation in vitro, apoptosis, cell invasion, migration and other biological behavior. Results Human telomerase reverse transcriptase RNA interference fragment could inhibit the activity of enzyme, inhibit the proliferation of T98G glioma cells (P <0.05), induce apoptosis (P <0.05) and inhibit the migration and invasion of cells (P <0.05). Conclusion Inhibition of human telomerase reverse transcriptase activity can effectively inhibit glioma cells in vitro biological behavior and promote apoptosis.
