目的观察雷公藤内酯醇对佐剂性关节炎大鼠外周血T淋巴细胞与趋化因子(MCP-1)趋化作用的影响。方法以佐剂性关节炎大鼠模型为研究对象,应用雷公藤内酯醇(0.2~0.4mg/kg,腹腔注射1次/d,共5d)后,分离大鼠外周血T淋巴细胞,观察MCP-1对T淋巴细胞趋化作用,检测病变关节肿胀度、外周血肿瘤坏死因子(TNF-α)含量。结果雷公藤内酯醇在0.2~0.4mg/kg范围内能明显减轻佐剂性关节炎病变关节肿胀度,减少外周血TNF-α含量,显著抑制MCP-1对T淋巴细胞趋化作用。结论雷公藤内酯醇具有抑制MCP-1对佐剂性关节炎大鼠外周血T淋巴细胞趋化作用的能力。
Objective To investigate the effects of triptolide on the chemotaxis of peripheral blood T lymphocytes and chemokines (MCP-1) in adjuvant arthritis rats. Methods The rat model of adjuvant arthritis was used as the research object. After the triptolide (0.2-0.4 mg / kg, intraperitoneal injection once per day for 5 days), T lymphocytes were isolated from peripheral blood of rats. MCP -1 on T lymphocyte chemotaxis, detection of diseased joint swelling, tumor necrosis factor (TNF-α) content in peripheral blood. Results In the range of 0.2-0.4 mg / kg, triptolide significantly reduced the degree of swelling in joints and decreased the level of TNF-α in peripheral blood and significantly inhibited the chemotactic effect of MCP-1 on T lymphocytes. Conclusion Triptolide has the ability to inhibit the chemotactic effect of MCP-1 on T lymphocytes in peripheral blood of adjuvant arthritis rats.
