目的由于Pax-2对肾细胞癌具有较高特异性,可以作为肾细胞癌的一种肿瘤标志物,因此探讨Pax-2在肾细胞癌患者外周血中的表达及其对微转移的意义。方法用RT-PCR法检测25例患者外周血中的Pax-2mRNA表达,以10例其他肿瘤患者和5例健康人为阴性对照,以β-actin为内对照。结果19例患者外周血中有Pax-2mRNA的表达(阳性率为76%),其他肿瘤和正常人中有1例表达Pax-2mR-NA(阳性率为6.7%),差异有显著性(χ2=18.027,P<0.01)。Pax-2mRNA的表达与肿瘤分类、临床分期无相关性(χ2=3.206,3.181,P>0.05)。结论外周血中Pax-2mRNA的检测可以较敏感地检测到血液中肾癌细胞,有助于早期诊断肾细胞癌及其微转移,从而改善患者的预后,提高生活质量。
Purpose Because Pax-2 is highly specific for renal cell carcinoma and can be used as a tumor marker for renal cell carcinoma, we investigated the expression of Pax-2 in peripheral blood of patients with renal cell carcinoma and its significance for micrometastasis. Methods The expression of Pax-2 mRNA in peripheral blood was detected by RT-PCR in 25 patients. The other 10 cancer patients and 5 healthy controls were negative controls and β-actin was used as control. Results The expression of Pax-2 mRNA in peripheral blood was positive in 19 patients (positive rate was 76%). Pax-2mR-NA was expressed in 1 of the other tumors and normal individuals (positive rate was 6.7%), with significant difference (χ2 = 18.027, P <0.01). There was no correlation between the expression of Pax-2mRNA and tumor classification and clinical stage (χ2 = 3.206,3.181, P> 0.05). Conclusion The detection of Pax-2 mRNA in peripheral blood can detect the renal cancer cells in blood more sensitively, and help to diagnose renal cell carcinoma and its micrometastases early, so as to improve the prognosis of patients and improve the quality of life.
