目的为了解多药耐药MDR1基因表达产物P糖蛋白(Pgp)表达与癌基因的关系,选择Pgp、p53、表皮生长因子受体(EGFR)三个在膀胱癌中较常表达的肿瘤指标,研究在膀胱移行细胞癌的表达情况、临床意义,并针对Pgp表达与突变p53蛋白、EGFR之间是否存在相关性进行探讨。方法采用免疫组化SP法,应用单克隆抗体JSB1、DO-7、31G7分别检测64例原发性膀胱移行细胞癌中Pgp、p53、EGFR的表达情况,并与17例正常膀胱粘膜进行对照。结果64例原发性膀胱移行细胞癌中Pgp、P53、EGFR的阳性表达率分别为67.18%,51.56%,68.5%,与对照组比较P<0.01。Pgp、EGFR过表达与膀胱癌的病理分级、分期和术后存活率均无关,而与膀胱癌的复发有关(P<0.05);突变p53蛋白的表达除与膀胱癌的病理分级无关外(P>0.05),与膀胱癌的病理分期、复发和术后存活率均有关(P<0.05)。相关性研究显示Pgp表达与突变p53蛋白、EGFR的表达密切相关(P<0.01)。生存分析表明,膀胱癌的病理分级、分期和突变p53蛋白的表达状态均与术后存活率有关(P<0.05)。结论(1)Pgp、突变p53蛋白、?
Objective To investigate the relationship between Pgp expression and oncogenes in multidrug resistance MDR1 gene and to choose the three tumor markers which are often expressed in bladder carcinoma, including Pgp, p53 and epidermal growth factor receptor (EGFR) To investigate the expression and clinical significance of bladder transitional cell carcinoma (BTCC) and to explore the relationship between Pgp expression and the mutation of p53 protein and EGFR. Methods Immunohistochemical SP method was used to detect the expression of Pgp, p53 and EGFR in 64 cases of primary bladder transitional cell carcinoma by using monoclonal antibodies JSB1, DO-7 and 31G7, respectively, and compared with 17 cases of normal bladder mucosa. Results The positive rates of Pgp, P53 and EGFR in 64 cases of primary bladder transitional cell carcinoma were 67.18%, 51.56% and 68.5%, respectively, P <0.01 compared with the control group. The overexpression of Pgp and EGFR was not related to the pathological grade, stage and postoperative survival rate of bladder cancer, but was related to the recurrence of bladder cancer (P <0.05). The expression of P53 and p53 was not associated with the pathological grade of bladder cancer (P> 0.05), which was related to the pathological stage, recurrence and postoperative survival rate of bladder cancer (P <0.05). Correlation studies showed that Pgp expression was closely related to the expression of mutant p53 protein and EGFR (P <0.01). Survival analysis showed that the pathological grade, stage and the expression of mutant p53 protein in bladder cancer were related to postoperative survival (P <0.05).
