目的:研究荨麻多糖对D-半乳糖致衰老模型小鼠的抗衰老作用。方法:将90只C57BL小鼠随机分为正常对照组、模型组、VE(100 mg/kg)阳性对照组及荨麻多糖高(200 mg/kg)、中(100 mg/kg)、低(50 mg/kg)剂量组6组,正常对照组每日皮下注射生理盐水(10 m L/kg),其余各组小鼠均每日皮下注射D-半乳糖(150 mg/kg)造模,同时6组小鼠分别灌胃相应药物,正常对照组与模型组灌胃等体积生理盐水。给药42 d后,称量体质量,进行八臂迷宫试验和游泳耐力测验。随后处死各组小鼠,采集小鼠的血清、肝、脑检测其中总蛋白含量,并用试剂盒检测肝、脑总抗氧化能力(T-AOC),总超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)活性及丙二醛(MDA)含量。结果:与模型组比较,荨麻多糖组小鼠体质量增长幅度较大,游泳耐力更好,迷宫通过时间缩短,错误次数减少,血清与肝、脑组织蛋白含量及肝、脑组织总抗氧化能力和SOD、GSH-Px活性显著升高,而肝、脑组织MDA含量显著降低。结论:荨麻多糖对D-半乳糖致衰老小鼠有一定的抗衰老作用,其机制可能与抗氧化作用有关。
Objective: To study the anti-aging effect of nettle polysaccharides on aging model mice induced by D-galactose. Methods: 90 C57BL mice were randomly divided into normal control group, model group, VE (100 mg / kg) positive control group and high net polysaccharide (200 mg / kg) 50 mg / kg). The normal control group was injected subcutaneously with normal saline (10 m L / kg). Results: Compared with the model group, the nettle polysaccharide group showed a significant increase in body weight, swimming endurance, maze passage time shortened, the number of errors, serum and liver, brain tissue protein content and liver, brain total antioxidant Ability and SOD, GSH-Px activity was significantly increased, while liver, brain MDA content was significantly reduced. Conclusion: Nettle polysaccharides have certain anti-aging effects on aging mice induced by D-galactose, and its mechanism may be related to anti-oxidation.
