目的建立手性分离-液相色谱/质谱法,用于分离检测血液中艾司佐匹克隆。方法血液经HLB小柱固相萃取后,采用手性分离柱,用电喷雾正离子模式离子化、多反应监测模式检测艾司佐匹克隆,外标法定量。结果采用本文方法可有效分离佐匹克隆的左旋、右旋异构体,血中艾司佐匹克隆在1~100ng/mL范围内线性关系良好,相关系数为0.996 4,最低检出限为0.8ng/mL,回收率为76%~88%,日内与日间精密度均小于8%。结论本文所建方法简便、快速、分离度好,适用于血液中艾司佐匹克隆的检测。
Objective To establish a chiral separation - liquid chromatography / mass spectrometry for the determination of eszopiclone in blood. Methods After the blood was solid-phase extraction with HLB cartridge, the chiral separation column was used. The ionization was performed by electrospray positive ion mode. The eszopiclone was detected by multiple reaction monitoring mode. Results The levodal and dextrorotatory isomers of zopiclone can be effectively separated by this method. The linearity of eszopiclone in blood ranged from 1 ng / mL to 100 ng / mL with a correlation coefficient of 0.996 4 and a minimum detectable limit of 0.8 ng / mL, the recovery was 76% ~ 88%, the intra-day and inter-day precision were less than 8%. Conclusion The method proposed in this paper is simple, rapid and has good resolution and is suitable for the determination of eszopiclone in blood.
