目的:观察哮喘宁颗粒药物含药血清对大鼠嗜碱性粒细胞株(RBL-2H3)细胞脱颗粒的影响,以探讨哮喘宁颗粒治疗支气管哮喘的疗效机制。方法:SD大鼠,均分5组,分别按哮喘宁颗粒0.4,0.2,0.1 g.kg-1、地塞米松(阳性对照)8.33×10-5g.kg-1、同体积生理盐水阴性对照,均ig,连续5 d,制备哮喘宁颗粒含药血清;以5%,10%含药血清干预RBL-2H3细胞脱颗粒进程,用钙离子成像法观察细胞脱颗粒早期细胞内钙离子的变化;用比色法检测细胞脱颗粒后β-己糖苷酶的释放量。结果:哮喘宁颗粒含药血清可以抑制Compound 48/80致敏的肥大细胞脱颗粒,在脱颗粒早期抑制细胞内钙离子的升高,并抑制β-己糖苷酶的释放。结论:哮喘宁颗粒含药血清在体外可以抑制致敏肥大细胞的脱颗粒过程,这可能是哮喘宁颗粒剂抑制哮喘I型变态反应,治疗支气管哮喘的机制之一。
Objective: To observe the effect of asthma ning granule drug-containing serum on degranulation of rat basophil cell line (RBL-2H3) and to explore the therapeutic mechanism of asthma ning granule in treating bronchial asthma. Results: Astragalus granule-containing serum could inhibit Compound 48/80-sensitized mast cells degranulation, inhibit the increase of intracellular calcium ion and inhibit the release of β-hexosaminidase at the early stage of degranulation. CONCLUSION: Asthma Ning Granule contains serum can inhibit degranulation of sensitized mast cells in vitro, which may be one of the mechanisms of Asthma Ning granules inhibiting type I allergic reaction of asthma and treating bronchial asthma.
