目的:研究树突状细胞(dendriticcels,DC)类瘤苗免疫小鼠脾淋巴细胞对实验性骨髓瘤的继承性免疫预防与免疫治疗的作用。方法:先前用骨髓DC作为佐剂,研制了DC与肿瘤细胞的融合瘤苗,以及用灭活肿瘤细胞及其裂解产物体外脉冲的DC疫苗,并成功地诱导宿主产生抗肿瘤的免疫保护性反应,且对荷瘤动物具有免疫治疗作用。在此基础上,本研究用DC融合瘤苗及肿瘤抗原脉冲DC疫苗免疫的脾淋巴细胞作为免疫效应细胞,继承性转输到宿主体内,以观察其免疫预防与治疗的效果。结果:继承性免疫预防试验表明,转输免疫脾淋巴细胞后2组宿主均能够抵抗野生性肿瘤细胞的多次攻击,>60d未见皮下移植性肿瘤形成,其60d存活率为100%(5/5),与对照组相比,P<0.01。继承性免疫治疗(adoptiveimmunotherapy,AIT)试验显示:①单用免疫淋巴细胞进行AIT的2组,60d存活率分别为80%(4/5)和60%(3/5),其存活时间也明显延长,P<0.05;②先行化疗后进行AIT,以及术前或术后再行AIT,60d存活率提高到100%(5/5),其存活时间显著延长,P<0.01;AIT后宿主荷瘤生长明显被抑制,且部分荷瘤消退甚或治?
OBJECTIVE: To study the role of splenic lymphocytes from mice immunized with dendritic cels (DC) vaccines against the inherited immune prevention and immunotherapy of experimental myeloma. METHODS: Previously, bone marrow DCs were used as adjuvants to prepare DC-tumor cell fusion vaccines and DC vaccines that were pulsed in vitro with inactivated tumor cells and their lysates, and successfully induced the host to produce an anti-tumor immune protective response. , And have immunotherapy for tumor-bearing animals.
