采用高效液相色谱法和放射性薄层层析法 ,检测了大鼠血小板和腹水白细胞花生四烯酸经脂氧合酶 (LO)代谢产物含量 ,探讨依布硒及其类似物α 甲基 4 ( 3 氧 2H 1,3 苯并异硒唑 2 基 )苯乙酸 (MB BA)对LO活性的抑制作用 .结果表明 ,依布硒 ,MBBA对 5 LO ,12 LO和 15 LO都有较强的剂量依赖性的抑制作用 ,特别是MBBA在 2 μmol·L- 1浓度时就可使花生四烯酸经 15 LO的代谢产物 15 羟基 ( 5,8,11,13) 二十碳四烯酸的含量减少 96% ,表现出强的抑制 15 LO的活性 .这种抑制作用对阻止低密度脂蛋白的氧化修饰有重要意义
The contents of lipoxygenase (LO) metabolites of arachidonic acid in rat platelets and ascites leucocytes were detected by high performance liquid chromatography and radioactive thin layer chromatography. The effects of ebselen and its analogues α methyl 4 (3 oxy-2H 1,3 benzoselenazole 2-yl) phenylacetic acid (MB BA) on the activity of LO were studied.The results showed that ebselen and MBBA had stronger LO, LO and LO than 5 LO Dose-dependent inhibition, in particular, MBBA at 2 μmol·L-1 concentration can make arachidonic acid by the 15 LO metabolites of 15 hydroxyl (5,8,11,13) arachidonic acid Content decreased by 96%, showing a strong inhibition of the activity of 15 LO.
