目的:探讨维甲酸(RA)诱导鼻咽癌端粒酶及hTR表达下降和凋亡改变。方法:10-5mol/L维甲酸处理HNE1细胞后,采用TRAP PCR ELISA、逆转录巢式PCR、凋亡指数及透射电镜检测端粒酶、hTR表达及凋亡改变。结果:加入维甲酸6 d后,HN E1细胞形态不规则,细胞悬浮,细胞数减少,端粒酶及hTR表达受抑制,间接定量显示端粒酶活性(0.68±0.14)U只及对照组(1.16±0.46)U的1/2,凋亡指数及细胞明显高于对照组(P<0.05)。结论:维甲酸能明显抑制鼻咽癌细胞中端粒酶及hTR表达,并诱导该类肿瘤细胞增殖受阻及发生凋亡改变。
AIM: To investigate the effect of retinoic acid (RA) on the decrease of telomerase and hTR expression in nasopharyngeal carcinoma (NPC) and the change of apoptosis. Methods: HNE1 cells were treated with 10-5 mol / L retinoic acid. TRAP PCR ELISA, reverse transcription nested PCR, apoptosis index and transmission electron microscopy were used to detect the expression of telomerase, hTR and apoptosis. RESULTS: After treated with retinoic acid for 6 days, the morphology of HN E1 cells was irregular, cell suspension and cell number were decreased, and the expression of telomerase and hTR was inhibited. The telomerase activity (0.68 ± 0.14) U was detected by indirect quantitation 1.16 ± 0.46) U 1/2, apoptosis index and cells were significantly higher than the control group (P <0.05). Conclusion: Retinoic acid can significantly inhibit the expression of telomerase and hTR in nasopharyngeal carcinoma cells and induce the inhibition of cell proliferation and apoptosis.
