目的 探讨ATP对噪声暴露听力损伤的保护作用及对耳蜗一氧化氮合酶活性的影响。方法 制备豚鼠稳态噪声暴露听力损伤模型 ;应用NADPH -d酶组织化学及免疫组化技术ABC法 ,结合听性脑干反应 (ABR)阈值检测 ,研究ATP对噪声暴露致聋豚鼠耳蜗一氧化氮合酶 (NOS)及诱生型一氧化氮合酶 (iNOS)活性的影响。结果 ATP对噪声暴露豚鼠的ABR阈移有明显的抑制作用 ;ATP对噪声暴露豚鼠耳蜗NOS、iNOS的活性升高有明显抑制作用。结论 ATP对噪声暴露所致听力损伤具有保护作用 ,这种作用可能与耳蜗NOS、iNOS活性降低有关
Objective To investigate the protective effect of ATP on noise-induced hearing impairment and its effect on the activity of nitric oxide synthase in the cochlea. Methods The hearing loss model of guinea pig exposed to steady-state noise was prepared. ATP was used to detect nitric oxide in cochleas of guinea pigs exposed to noise with NADPH-d enzyme-linked immunosorbent assay (ABC method) and auditory brainstem response (ABR) Synthase (NOS) and inducible nitric oxide synthase (iNOS) activity. Results ATP significantly inhibited the ABR threshold shift in guinea pigs exposed to noise. ATP significantly inhibited the activation of NOS and iNOS in guinea pigs exposed to noise. Conclusions ATP has a protective effect on hearing impairment caused by noise exposure, which may be related to the decrease of NOS and iNOS activity in cochlea
