目的:观察一氧化氮(NO)供体亚硝基铁氰化钠(SNP)作用正常精子后超微结构的变化。方法:采用percoll梯度离心法选择具有正常生理功能的精子,与不同浓度的SNP孵育作用后,在透射电镜下观察精子的超微结构。结果:正常精子与300nmol/L SNP作用后,精子膜、顶体及精子线粒体出现不同程度的损伤和结构上的异常。结论:高浓度NO供体SNP可致精子膜、顶体及精子线粒体等超微结构的改变,进一步阐明了精子运动功能障碍的机制。
Objective: To observe the ultrastructural changes of normal sperms after nitric oxide (NO) donor nitrosylated sodium cyanide (SNP). Methods: Percoll gradient centrifugation was used to select sperm with normal physiological function. After incubating with different concentrations of SNP, the ultrastructure of sperm was observed under transmission electron microscope. Results: Sperm membrane, acrosome and sperm mitochondria showed different degrees of damage and structural abnormalities after treated with 300 nmol / L SNP. CONCLUSIONS: High concentrations of NO donor SNP can induce ultrastructural changes such as sperm membrane, acrosome and sperm mitochondria, further elucidating the mechanism of sperm motility dysfunction.
