目的探讨人乳腺癌及正常乳腺组织中P-AKT的表达及其意义,为乳腺癌的早期临床诊断和治疗提供理论和实验依据。方法采用Western blot及RT-PCR方法检测100份乳腺癌组织标本和30份正常乳腺组织标本中P-AKT蛋白及mRNA的表达情况,并进一步探讨其与乳腺癌临床病理指标的相关性。结果 P-AKT蛋白及mRNA在乳腺癌中的表达水平高于在正常乳腺组织中的表达水平,且P-AKT蛋白及mRNA的表达与临床分期相关(P<0.05),而与年龄、肿瘤直径、淋巴结转移和组织学分级无关(P>0.05)。结论 PI3K/AKT信号通路可能参与乳腺癌的发生发展,在癌变过程中起着重要作用。
Objective To investigate the expression and significance of P-AKT in human breast cancer and normal breast tissues, and to provide theoretical and experimental evidences for the early clinical diagnosis and treatment of breast cancer. Methods The expression of P-AKT protein and mRNA in 100 specimens of breast cancer tissues and 30 specimens of normal breast tissues were detected by Western blot and RT-PCR. The correlation between P-AKT protein and mRNA and clinicopathological parameters of breast cancer was further explored. Results The expression of P-AKT protein and mRNA in breast cancer was higher than that in normal breast tissue. The expression of P-AKT protein and mRNA was correlated with clinical stage (P <0.05), but not with age, tumor diameter , Lymph node metastasis and histological grade (P> 0.05). Conclusion PI3K / AKT signaling pathway may play an important role in carcinogenesis and progression of breast cancer.
