目的观察白薇提取物对部分肝切除后血管再生过程的影响,探讨其调节作用的相关分子及机制。方法昆明种小鼠20只,随机分为对照组、白薇组。两组分别采用一次性切除部分肝左外叶。实验组则在术后开始灌胃给药白薇提取液(50 g/kg),2次/d。术后1~8 d,每天处死2组小鼠各1只,取肝左外叶。免疫组织化学技术检测血管内皮生长因子C(VEGF-C)、VEGF-A、淋巴管内皮透明质酸受体1(LYVE-1)、CD34的表达。结果与对照组相比,白薇组中CD34表达明显升高,分布范围扩大,VEGF-C、VEGF-A、LYVE-1的表达早期下降,后期则显著增加。结论白薇上调VEGF-A、VEGF-C、LYVE-1的表达,促进血管新生。
Objective To observe the effect of extract of Baiwei on the process of vascular regeneration after partial hepatectomy and to explore the related molecules and mechanisms of its regulation. Methods Kunming mice 20 were randomly divided into control group, Baiwei group. Two groups were used to remove part of the left hepatic lobe. In the experimental group, Bai Wei extract (50 g / kg) was given intragastrically once a day for 2 times / d. One to eight days after operation, 2 mice in each group were sacrificed, and the left outer lobe of the liver was taken. The expressions of VEGF-C, VEGF-A, LYVE-1 and CD34 were detected by immunohistochemistry. Results Compared with the control group, the expression of CD34 in the Baiwei group was significantly increased and the distribution range was expanded. The expression of VEGF-C, VEGF-A and LYVE-1 decreased in the early stage and then increased significantly in the later stage. Conclusion Bai Wei upregulates the expression of VEGF-A, VEGF-C and LYVE-1, and promotes angiogenesis.
