目的研究健脾复方对人大肠癌移植瘤裸小鼠血管内皮生长因子(vascular endothelial growth factor,VEGF)和肿瘤微血管密度(microvessel density,MVD)的影响。方法对60只BALB/C裸小鼠建立人大肠癌原位移植瘤模型,并按1:1:1:1:1随机分为健脾复方+5-Fu组、健脾复方组、塞来昔布组、5-Fu组和模型对照组,分别予相应的干预措施处理8周。分别观察VEGFmRNA(采用RT-PCR法)和MVD(免疫组化法)的表达,并对影响其表达的相关因素进行分析。结果与模型组比较,健脾复方+5-Fu、健脾复方、塞来昔布和5-Fu的治疗均可降低VEGFmRNA的表达(P<0.01),健脾复方+5-Fu、健脾复方可进一步降低MVD表达,且与5-Fu组和模型对照组的差异有显著性(P<0.05)。回归分析显示VEGFmRNA和MVD的表达与不同的治疗方案具有相关性。结论健脾复方单独使用或联合5-Fu,均可显著抑制大肠癌瘤块VEGFmRNA和MVD的表达,提示健脾复方有抑制肿瘤生长、复发、转移的潜能。
Objective To study the effect of Jianpi Compound on vascular endothelial growth factor (VEGF) and microvessel density (MVD) in human colorectal cancer xenografts. Methods 60 human BALB / C nude mice were established in situ xenograft model of human colorectal cancer and were randomly divided into five groups according to the ratio of 1: 1: 1: 1: 1. Cox group, 5-Fu group and model control group were given corresponding interventions for 8 weeks. The expression of VEGF mRNA (using RT-PCR method) and MVD (immunohistochemistry) were observed, and the related factors that affect the expression were analyzed. Results Compared with the model group, the treatment with spleen-invigorating compound + 5-Fu, spleen-invigorating compound, celecoxib and 5-Fu both could reduce the expression of VEGFmRNA (P <0.01) Compound can further reduce the expression of MVD, and 5-Fu group and model control group differences were significant (P <0.05). Regression analysis showed that the expression of VEGF mRNA and MVD correlated with different treatment regimens. Conclusion Spleen-invigorating compound alone or in combination with 5-Fu can significantly inhibit the expression of VEGFmRNA and MVD in colorectal cancer, suggesting that the spleen-invigorating compound can inhibit the potential of tumor growth, recurrence and metastasis.
