目的探讨<3岁反复喘息患儿外周血血清类黏蛋白1样蛋白3(ORMDL3)基因在不同临床表型中的表达及其与血清白介素-17(IL-17)和嗜酸性细胞阳离子蛋白(ECP)的关系。方法选取该院儿科就诊<3岁反复哮喘的患儿87例,根据患儿是否存在高危因素分成特应性哮喘组(64例)和非特应性哮喘组(23例),再选取同期在儿科体检的健康儿童30例。分析3组研究对象ORMDL3基因表达水平、血清IL-17和ECP含量和ORMDL3基因频率分布的差异,并对3组研究对象ORMDL3基因表达水平和血清IL-17和ECP含量的相关性进行分析。结果特应性哮喘组ORMDL3基因表达水平(11.57±1.96)%,血清IL-17(1 354.46±873.48)pg/ml和血清ECP(45.65±12.76)μg/L均高于非特应性哮喘组和对照组(P=0.000);3组的ORMDL3基因AA、AG和GG 3种基因型的频率分布差异有统计学意义(χ~2=12.84,P=0.012)。特应性哮喘组的ORMDL3基因表达水平与血清IL-17(r=0.318,P=0.000)、血清ECP(r=0.540,P=0.000)均呈正相关。结论不同临床类型的哮喘患儿血清中的ORMDL3基因表达水平、IL-17和ECP含量不同。特应性哮喘患儿的血清ORMDL3基因表达水平与血清IL-17、ECP含量存在正相关性。
Objective To investigate the expression of ORMDL3 gene in children with recurrent wheezing at 3 years old in different clinical phenotypes and its relationship with serum interleukin-17 (IL-17) and eosinophil cationic protein ECP) relationship. Methods Eighty-seven children with pediatric asthma <3 years of age were divided into atopic asthma group (64 cases) and non-atopic asthma group (23 cases) according to their risk factors. Physical examination of 30 healthy children. The ORMDL3 gene expression levels, serum IL-17 and ECP levels and ORMDL3 gene frequency distribution in the three groups were analyzed. The correlation between ORMDL3 gene expression levels and serum IL-17 and ECP levels was also analyzed. Results The ORMDL3 gene expression level (11.57 ± 1.96)%, serum IL-17 (1 354.46 ± 873.48) pg / ml and serum ECP (45.65 ± 12.76) μg / L in atopic asthma group were significantly higher than those in non-atopic asthma group (P = 0.000). There was significant difference in the frequency distribution of ORMDL3 gene AA, AG and GG between the three groups (χ ~ 2 = 12.84, P = 0.012). ORMDL3 gene expression in atopic asthma group was positively correlated with serum IL-17 (r = 0.318, P = 0.000) and serum ECP (r = 0.540, P = 0.000) Conclusions Serum levels of ORMDL3 in children with different clinical types of asthma are different, with different levels of IL-17 and ECP. Serum ORMDL3 gene expression levels in children with atopic asthma have a positive correlation with serum IL-17 and ECP levels.
